We examined Cortisol (F) dynamics in female baboons treated with diethylstilbestrol (DES) or estradiol (E2) and compared values with those previously measured in nonpregnant and pregnant animals. Five regularly menstruating baboons (12–18 kg, BW) were administered 5 mg DES daily via fruit or 0.5 mg E2/0.1ml oil sc for 30 days. Blood samples, obtained before and after treatment, were assayed for serum F concentrations and serum Cortisol binding capacity (CBC). The metabolic clearance (MCR) and production rate (PR) of F and the catabolism of i.v. administered [3H] F were examined 25 and 30 days after initiation of estrogen treatment. Compared with values in nonpregnant baboons, F metabolism in estrogen treated animals is significantly altered and is characterized by increased formation of unconjugated metabolites, decreased glucuronylation, increased excretion of unconjugated F, cortisone, and highly polar metabolites, and increased CBC. These changes induced by estrogen are similar to those observed in intact pregnant baboons and permit the suggestion that the pattern of F metabolism and the level of CBC in baboon pregnancy are the result of elevated estrogen production.However, estrogen also caused a significant decrease in the MCR and PR of F, parameters which, by contrast, are similar in intact pregnant and nonpregnant baboons. These findings indicate that while estrogen also influences the rate of F clearance and F production, these effects of estrogen are not apparent during pregnancy. Collectively, these findings allow the suggestion that estrogen is a major factor which alters F metabolism and increases serum CBC in baboon gestation. However, additional factors are operative in primate pregnancy which maintain PR and MCR of F at levels similar to those of nonpregnant baboons. 相似文献

Potassium (KCl: 50 mM) increases the uptake of 45Ca by GH3/B6 pituitary cells whereas TRH (100 nM) does not modify the uptake. On the contrary, 45Ca efflux is quickly and transiently increased by TRH whereas KCl has no influence on the 45Ca outflow. Quantitative locomotion changes have been studied in the norm and in different periods after local ablation of motor projection of hind paw in the right hemisphere of 5-16 weeks white rats. The length and width of step gradually increase with the age, and coefficients of gait asymmetry reflecting individual characteristics, are relatively stable in intact animals. Local decortication is accompanied by significant shifts of all locomotion parameters clearly expressed in the first days after ablation. The most sensitive characteristic of gait anomaly is the standard deviation of half-step. In 5 weeks after ablation a lag is observed of operated rats behind the control ones in all parameters. Load application contributes to revealing of locomotion parameters shifts from the norm in later periods after decortication. In 9 and 11 weeks after surgery, the signs of supercompensation are observed in a number of parameters. 相似文献

Adenylate cyclase activity was measured in broken cell preparations of whole endometrial tissue from rabbits on Days 0, 1, 6.5, 9 and 15 of pseudopregnancy and in endometrial epithelial and stromal cells on Days 1 and 6.5 to assess the specific response of individual cell types. In dispersed cells, adenylate cyclase activity was higher (P less than 0.01) in stromal than in epithelial cells and reduced on Day 6.5 compared to Day 1 in both cell types. The response of adenylate cyclase to isoproterenol appeared more important relative to the PGE-2 response in epithelial than in stromal cells and strongly reduced in the former on Day 6.5. In endometrium, the overall adenylate cyclase activity was increased significantly on Day 1 of pseudopregnancy compared to Day 0 (oestrus), only 18 h after injection of hCG. On the following days, the activity decreased progressively on Days 6.5 and 9 and exhibited a recovery on Day 15. Adenylate cyclase response to isoproterenol (% over GTP) was comparable on Days 0, 1 and 6.5, abolished on Day 9 and recovered on Day 15. Maximal response to PGE-2 (% over GTP) was observed on Day 6.5, at the time of implantation, maintained on Day 9 and reduced on Day 15 towards the low levels measured in oestrus and Day 1 of pseudopregnancy. Our results demonstrate a dramatic alteration of adenylate cyclase activity in rabbit endometrium during pseudopregnancy. It suggests a possible involvement of catecholamines and prostaglandin E-2 in the regulation of endometrial receptivity through a cAMP-mediated process. 相似文献

Preparations of ColEl plasmid DNA synthesized in the presence of chloramphenicol were separated into samples having gaps resulting from removal of ribonucleotides in one or the other of the complementary DNA strands. These samples were used as templates for repair resynthesis reactions using DNA polymerase of Rous sarcoma virus and α-32P-labeled deoxyribonucleoside 5′-triphosphates. Reactions involved the incorporation of each labeled nucleotide in the presence of three unlabeled nucleotides, and also the incorporation of all four labeled nucleotides followed by complete digestion and electrophoretic separation of the products. By these two methods the RNA integrated in the light strand of ColEl DNA was found to comprise an average of 38 ribonucleotides with a base composition of 17G, 5A, 8C, and 8U. The RNA segment in the heavy strand consists of an average of 15 ribonucleotides with a base composition of 5G, 2A, 4C, and 4U. 相似文献
